I'm only familiar with TStreet. Until this past August, I was an active reviewer, till the poor quality of the SPs started to make me physically ill.
The quality of feedback you get on this site is wildly variable. Some of the reviewers give high-quality development notes; others are trolls who keenly attack whoever ends up on their review list. And there's some fluff in between.
If a reviewer trashes you or gives unconstructive criticism, you have the option of reporting the matter to the Hall of Justice. The message boards contain some serious advice.

I'm a member of both. Zoetrope is a bit more 'artsy.'
If you're a beginner it's useful to do reviews and learn. The reviews vary wildly in quality. Might be better to get pro feedback. A few guys listed on DoneDealPro boards ('consultants' thread). EvilRobot/Andrew is good and cost 75 bucks, less if you don't want the synopsis.
There's a new website -- Talentville.com. Similar thing.

It's been a while since I've used either, but I think both are useful for an early draft. This is your potential audience. Most aren't professional readers or writers and they look at the script differently. You can get some very skilled people and you can request that a particular reader on Zoetrope reads your script. You can trade reads. You also will get some very bizzarre or useless comments. You need to be skilled enough to know if it's something you can use or not.
